R&D department will be running peanut butter as a trial run on one line. what kind of prep work or isolation should be used for nearby line?

I would run the trial POST operation and pre sanitation on the adjoining lines and then swab to ensure there is no peanut protein present prior to running non peanut again (assuming you're not running peanut at all currently)

I agree with Scampi.
Do a full risk assessment in advance with the HACCP-team. You will find many points to be considered. We habe a special focus on peanuts.
But it depends on the industry (we are a bakery with many allergens) and the kind of equipment involved (old or new -> hygenic design etc.). In any cases you should combine this with the general cleaning validation of the line and all things/equipments being involved (rework, cleaning and handling equipment, clothes of workers and...and...). We are using special colour coding for allergens and a special one for peanut, but in the frist trial situation separation is sufficient to separate. Keep an eye on the washing equiment and the recycled water..and...and..).

Make sure incoming ingredient(s) that contain the peanut allergen are properly labeled, stored and removed from the facility after the trial, and employees are prepped on handling these materials. Also to consider is storage of the trial products. Cleaning tools that are used should also be included in a swab test as Scampi suggested for the lines.
