Method for Salmonella / 250 gm

Usually in a food sample Salmonella is tested per 25 gm ,However we have requirement to report it Salmonella /250 gm
kindly share method/protocol Or the changes we will have to make in the methods usually we follow BAM

I believe it would be the enumeration of 10x 25gm samples and then reported as enumeration of 250 gm

If 250gm is used and composited, probably need to adjust the volume of enrichment broth as detailed in BAM, eg -
1) The number and weights of the sample units is correct.
Each sample should be mixed to ensure homogeneity before withdrawing a 25 g analytical unit. The analytical units can be composited (fifteen 25 g units into a 375 g composite), unless otherwise indicated in Chapter 5 or in the OMA. Samples should be preenriched at a 1:9 sample-to-broth ratio.
(one common problem is lack of suitable size containers).
Alternatively, do individual x 10.
We are also asked to run Salmonella in bulk quantities (375g is our usual request) as well. For 250g, you can run 10 individual samples, or you can make the 1:10 dilution with 250 g sample to 2250ml of buffer. This way you only have to transfer/streak the one sample. We currently send the sample out to Covance Laboratories to run this bulk testing. We only run it 8 or 10 times a year, and they only have a small up-charge to run the larger volume. We have them run a method suitability once per product and then we have proof the method works. Still cheaper and a better use of our Microbiologists time than running 10 (or in our case 15) separate Salmonella samples.
