Is a dedicated COP Room required for carrying out manual cleaning-out-of-process? Would it be feasible to perform this task in 3 tier sink located in batching area or is the contamination risk too high?
Any insight would be greatly appreciated.

Enclosed area, yes - open area - no and that is for as you stated - contamination.

The real answer..."it depends". You have to assess the risk of having a 3 compartment sink in the batching area.
For example, if you are doing sanitation in the batching area while using the 3 compartment sink then the risk is low. But, carrying out the COP cleaning while in production in batching area that depends on the proximity and exposure which only you can assess.
