Hi everyone. I'm looking for standard operationg procedure for environmental swab test for e-coli, staphylococcus aureus and coliform.

Hope this procedure may help you.

SOP FOR COLLECTION OF SWAB SAMPLE

Objective:

To lay down the procedure for collection of swabs sample from surface of equipment’s and floor

Scope:

This SOP shall be applicable to all the operational areas which are in contact with the product

Responsibility:

Microbiologist is responsible to implement this document.

Accountability:

Quality Assurance Manager.

Requirements:

ü  Sterile swabs

ü  Normal Saline or phosphate buffer solution

Procedure:

ü  Take a sterile and sealed cotton swab,

ü  Add 2-3 ml phosphate buffer saline having pH 7.2 or Normal saline (Sterilized),

ü  Take the sterilized wet swab to the sampling area,

ü  Wear hand gloves, take out the swab from the tube and swab the surface of the equipment/floor, covering an area of 10x10 sq.cm, in unidirectional movements but not to and fro movements,

ü  After swabbing place the swab in tube containing normal saline or phosphate buffer saline

ü  Shake the tube gently

ü  Label the swab with appropriate sampling position and date.

ü  Transfer the swab to the lab immediately,

ü    Swab sample should be tested within 24 hours.

Acceptable limits:

After cleaning acceptable limits 

TVC  <100 cfu/cm2

E.coli <10 cfu/cm2

Staph <100 cu/cm2
